| I. Background | |

SIRPα（Signal Regulatory Protein α）是一种免疫球蛋白超家族跨膜蛋白，主要表达于 髓系细胞（如巨噬细胞、树突状细胞）表面，其胞外区含三个 Ig 样结构域（D1、D2、 D3），其中 D1 结构域特异性结合 CD47；胞内段含免疫受体酪氨酸抑制基序（ITIM）， 可招募 SHP-1/SHP-2 磷酸酶传递抑制信号。SIRPα顺利获得调控吞噬活性维持自身免疫耐受， 防止巨噬细胞误伤正常细胞。
CD47 是一种五次跨膜蛋白，广泛表达于红细胞、血小板及肿瘤细胞表面，其胞外 N 端 IgV 样结构域与 SIRPα结合，被称为“别吃我”（Don’t eat me）信号分子，在红细胞稳 态（防止脾脏巨噬细胞清除年轻红细胞）及肿瘤免疫逃逸中起关键作用。 当 CD47 结合 SIRPα后，SIRPα胞内 ITIM 基序被磷酸化，招募 SHP-1/SHP-2 磷酸酶， 顺利获得去磷酸化作用抑制下游促吞噬信号。 |
